Detection of PAI-1 4G/5G polymorphism

PAI-1 (Plasminogen Activator Inhibitor 1) is a 52 kDa glycoprotein. It is the main inhibitor of activators of plasminogen, and hence fibrinolysis and blood clot degradation. PAI-1 gene is located on chromosome 7 and consists of 9 exons. It includes many variations. The most significant variation, however, lies in the promoter region at -675, which is a single guanine base insertion/deletion known as 4G/5G polymorphism. While 5G allele binds both transcription activator and repressor, 4G allele only binds activator and leads to higher expression of PAI-1. 4G allele is considered a predisposing factor and has been related to various clinically important conditions including deep vein thrombosis, atherosclerosis and myocardial infarction, cancer, sepsis, miscarriage and asthma
